I personally have not yet tried the multiplayer modes, but reviews on ProtonDB seem positive. These were easily remedied by restarting the game. I've got 61 hours clocked in the game on Solus Budgie Linux with a 980ti and an X-series i7, and the only issues I've seen were occasional audio and framerate degradation after long play sessions. So to answer your question, yes, the game works beautifully in Proton, but it takes a couple steps to run after first install, and again after each Larian update. I saw some old threads asking about how the Definitive Edition plays on Linux, but nothing recent, and only one comment four months ago which links to the ProtonDB page saying how to get the game working in Steam Play.
